:
The gamma polyglutamic acid (γ-PGA) environment-friendly material is soluble in water, biodegradable, edible, non-toxic and non-induced immune system. This acid and its derivatives have many interests in multiple areas, including sewage treatment, food production and can also be adjusted ancestry carboxylic side chemically to produce many pharmaceutical compounds or to improve the qualities of the polymer product. γ-PGA is a form of natural substances produced by many Gram positive microorganisms. Twenty five bacterial isolates of microorganisms were screened for γ-PGA production with medium containing L-glutamic acid as the only nitrogen source. All isolates were obtained from samples of soil and water, collected from different places of Jeddah, Saudi Arabia. The production of γ-PGA in bacterial isolates with different taxonomical properties was investigated. The amount of γ-PGA was determined by measuring the medium viscosity. Seven isolates out of 25 (28%) of the tested isolates produced γ-PGA. The highest γ-PGA producer was the isolate (TM13) thus; it was selected for more detail studies. Based on its morphological, biochemical and physiological characteristics, as well as 16S rRNA sequence and phylogenetic analysis, this isolate was identified as Bacillus licheniformis TM13. The effects of different factors on the polymer production were studied. It was showed that broth medium E was the most favorable one, the optimum temperature was at 37°C, and incubation period required for maximum production was 6 day with aeration level at 150 rpm and pH ranged from 6-8. Different carbon sources and nitrogen sources were used for bacterial growth and γ- PGA production. Glucose proved to be the best carbon source, yeast extract was found to be the best nitrogen source. Moreover, γ-PGA was purified and characterized using SDS-PAGE, mass spectrometry and thin layer chromatography. All analysis indicated that γ-PGA is a homopolymer composed of D and L glutamic acid. ii
